The study involves the gut microbiome, which contains about 100 trillion bacteria, and the promotion or prevention of the effects of these microbes is an emerging field. Scientists at Einstein College of Medicine found that a specific bacterial byproduct has a crucial role in the intestinal tract. It is used to maintain the intestinal epithelial single cells that keep enteric bacteria and toxins in the intestine and will not be absorbed into the human body. Some diseases are caused by the breakdown of the intestinal epithelium.
"These bacteria and their metabolites affect the integrity of the intestinal epithelial cells, but how they work is not yet known," said Dr. Sridhar Mani, MD. Mani and his colleagues suspect that bacterial metabolites bind to receptors (PXRs) of intestinal epithelial cells to activate proteins and exert their effects. In a series of mouse experiments, the researchers found that a metabolite known as å²å“š3-propionic acid (IPA), through the activation of PXR, helps digestion and enhances the barrier function of the intestinal epithelium and prevents inflammation. More specifically, activation of PXR inhibits the production of TNF-[alpha], while enhancing binding proteins between adjacent intestinal epithelial cells.
"With the addition of IPA-producing bacteria or direct management of IPA, we may be able to prevent or treat inflammatory diseases caused by damage to the intestinal epithelium," said Dr. Mani. "Such a strategy can also be tried on some other forms of health issues including liver disease, diabetes, asthma, allergies, obesity and heart disease." Now Mani's team is developing new probiotics that are designed to interact via PXR Role, restore the barrier function of intestinal epithelial cells.
